Ageing and many human diseases are associated with oxidative stress, which is a condition reflected by a loss of redox signaling and control resulting in irreversible cellular damage by reactive oxygen/nitrogen species. Despite the wide use of biomarkers of oxidative damage in clinical research, accurate quantification of oxidative stress in vivo remains elusive. In this presentation, new strategies for artifact-free assessment of oxidative stress will be presented based on targeted and non-targeted metabolite profiling (i.e., metabolomics) of biological fluids using capillary electrophoresis-mass spectrometry (CE-MS). An introduction of CE/CE-MS technology and metabolomics will first be presented in the context of successful expanded newborn screening programs for early detection of inborn errors of metabolism [1, 2]. Recent strategies for artifact-free analysis of thiols will then be discussed when using CE-MS for determination of cellular redox status and antioxidant capacity based on the differential rates of glutathione oxidation [3]. A differential metabolomics study using CE-MS in conjunction with multivariate data analysis will also be presented in order to assess the efficacy of nutritional intervention to attenuate exercise-induced oxidative stress on a healthy volunteer [4]. The development of reliable analytical methodologies for metabolomics is critical towards improved understanding of the causal relationship between oxidative stress and human health.
